Abstract : RNA editing is a vital process of post-transcriptional modification that occurs widely occurs in plant organelles. Although, RNA editing methodical studies have been conducted in various plants, very few information is available on Catharanthus roseus. In the present study, editing events in photosynthesis complexes were examined and compared among different tissues, flower, leaves, stems, roots, hairy roots, and seedling of C. roseus. Transcriptome data of C. roseus plastome genes and RNA-Seq were downloaded from the National Center for Biotechnology Information (NCBI) and reference was assembled using bioinformatic tool. In total 80 C-to-U editing sites were identified including 35 non-synonymous and 45 synonymous edited sites. Leaves and stems tissues have the most frequent editing sites with 65 and 66 edits respectively, followed by seedlings with 64 edits of the total edited sites. By contrast, some of the editing sites were significantly reduced in the remaining tissues; flowers (59), roots (59) and hairy roots (55). A comparison of C. roseus editing sites of 18 plant species led to discovery of 50 novel editing sites in 11 genes. Furthermore, affected amino acids were analyzed and compared among the six different tissues. These analyses exhibited that RNA editing increased the hydrophobicity of the total converted amino acids for approximately 82%. The findings show that RNA editing in plastid plastome of C. roseus is tissue dependent and differentially regulated in different types of tissues.
