Abstract : The aim of current study was to isolate and identify major constituents of periploca somaliensis fruits (family Asclepiadaceae) and assess their cytotoxic activity on three different cell lines: MCF-7 (breast cancer), HepG2 (hepatocellular carcinoma), and HCT-116 (colon cancer). Chemical investigation of CHCl3 fraction of P. somaliensis afforded a new scalarane sesterterpene, namely perisomalien A (1), along with lupeol acetate (2), β-amyrin (3), cycloart-23Z-ene-3b,25-diol (4), and β-sitosterol-3-O-β-D-glucopyranoside (5). Their structures were elucidated by extensive spectroscopic methods including 1D (1H and 13C NMR) and 2D NMR (1H-1H COSY, HSQC, HMBC, and NOESY), in addition to comparison with literature data. Compound (4) showed relatively potent cytotoxic activity on MCF-7 with IC50 9.0 µM, compared to remaining compounds that had moderate effects with IC50 ranging from 19-24 µM. it is worthy noted that all compounds were less cytotoxic towards the other cell lines HepG-2 and HCT-166, with compound (4) possessing the lowest IC50 (25.9 and 19.9 µM) on HepG-2 and HCT-166, respectively.
