Abstract Rosa damascena Mill. is a member of the genus Rosa L., which belongs to the family Rosaceae. In Saudi Arabia, this species is growing mainly in Taif and Almadinah cities. According to local people, there are some differences between Rosa damascena grown in Taif and those grown in Almadinah. Taif Rose is used mainly for oil and water extraction, while Almadinah Rose is used for ornamentation and drinking with tea. Since there is no available scientific information about the differences between both Roses, this work was aimed to find more morphological, phytochemical and genetical details regarding to the differences between both Roses in order to reserve this species. In morphological studies the characteristics of both plants were examined in the field and in herbarium. There were some differences in the appearance. The distribution and shape of spines and prickles on the stem, the shape and size of the leaves, the size of the pedicel and gynoecium of flowers were the most useful treats. In chemical content, leaf extracts were examined from each plant by paper chromatography, thin layer chromatography and highperformance liquid chromatography. This revealed that some compounds such as Quercetin-3Rhamnoside,Quercetin-3-monorhamnoside, Myricetin-3-monorhamnoside, Gossypetin-7monoglucoside and Kaempferol-3-monoglucuronide were detected in Taif Rose , whereas they were undetected in Almadinah Rose . In addition, the quantity of quercetin in Almadinah Rose is higher than Taif Rose with ratio Almadinah/Taif = 4.58.
